Quantitative PCR probes - TaqManTM Probes
Now also available with Black Hole Quenchers - BHQ
TaqManTM probes are oligonucleotide probes that contains a fluorophore (5'-Fam or others) and a quencher (3'-Tamra or 3'-BHQ). As long as the fluorophore and the quencher are linked to the oligonucleotide, the fluorescence is quenched. During the amplification reaction, the 5' => 3' exonuclease activity of the Taq polymerase cleaves the fluorophore from the probe. Since the fluorophore is no longer in close proximity of the quencher, the fluorescence is emitted and can be measured. The fluorescence is in direct proportion to the amount of target DNA accumulation during the PCR cycles. TAG Copenhagen recommends the probes to be 20-29 bases long.
TaqManTM oligos from TAG Copenhagen are always RP-HPLC purified and quality controlled by MALDI-TOF MassSpec.

Degenerated oligos - oligos containing mixed bases
TAG Copenhagen do not charge extra1 for mixed bases (wobbles) when they are in equal representation (25% of each base, 33% of 3 bases or 50/50% of 2 bases).
When ordering oligos containing wobbles please use the following codes IUPAC-IUB symbols:
R (A or G), Y (C or T), M
(A or C), K (G or T), S (G or C), W (A
or T),
H (A or C or T), B (G or T or C), V
(G or C or A), D (G or T or A), N (G or A or T or C).

TAG Copenhagen also offers doped oligos with mixed bases in un-equal representation. For best result please limit the number of different mixed positions to 40 and limit the total sequence to 100 bases.
