    PTO oligos are known to be up to 100 times more resistant against intracelluar nucleases than unmodified oligos. Their specific translation inhibition can be very effective.

    The PTO oligos are purified by a combined RP-HPLC and desalting method. The crude PTO oligo is injected into a column and using gradients of acetonitrile and buffer the product is separated according to the size and polarity of the molecules. The main peak is detected by an online computer and collected using a robot. The collected product is deblocked and 100% desalted on a special second column.
